24-hour time-lapse videos, started ~5 mins after cell seeding of BMSCs cultivated on either thin (~50 µm; left) or thick (~200 µm; right) low elastic modulus, soft, PAAm substrates. Phase contrast videos of cells are shown in the top panels and fluorescence videos on bead displacement are shown in the bottom panels. Although pre-existing wrinkles are evident only on thick PAAm hydrogels, cells rapidly deform them to create cell-contraction induced wrinkles. Note also qualitatively that displacements are larger on thick PAAm hydrogels.
